I suspect it's something more like the inn was built on a sealed goddess of yuri that infects anyone that stays there to the point that even when het girls stay, they suddenly turn gay, like the first couple.

Only one sealed goddess? Wouldn't it make more sense that it would be two goddesses -- who happen to be each other's lover and maybe even sister! -- that are sealed on the grounds of the inn? Heck, why even sealed? Maybe the Kagaya Inn lies adjacent to or actually contains a shrine to a duo of yuri goddesses!

Kagaya Inn is the inheritance of a long line of sister-wife couples and their daughters who have run the inn for generations. The spirits of the Kagaya clan watch over the property and guide young ladies, both their descendants and the visitors who come there, into the delights of yuri.
How it comes to be that there are multiple generations, and how it is that the family line seems to produce only daughters, are closely guarded secrets passed down from mother to daughter along with the sacred Kagaya treasures, the turkey baster and the coat hanger. ...okay, that's a bit too warped. By what the strip itself shows, however, it seems that they're either bi and capable of producing children while having their yuri on the side, or else they are straight and reproduce while young, then, when they inherit the inn themselves, they come back and get converted by the yuri field.
Looks just as likely to be an androgynous (and possibly tomboyish) girl to me. Haruka Ten'oh, anyone?
