Our best understanding of "sleep" right now is that the brain, like all other organs and cells, produces and accumulates waste product over time. "Sleep" is how it safely flushes it out without disrupting anything important.
So, as you can imagine, a lack of sleep means a literally crappy brain.

Which translates to dementia and brain damage over time. Your body loses its command center, and your immunity drops too, leading to increased risk of disease.
There are a number of studies that also link sleep to heart health. The above listed effects can result in hypertension and heart illness as well. Heart issues in particular can also be exasperated by abusing caffeine and energy formula while low on sleep.
Ultimately, there is no suitable substitute for sleep for those reasons.
