Think RJ's breast envy was also purely fanon, with canon RJ being proud of her "unique silhouette". So, kinda like Tama.

...Though, moving back to Umamusume, think they also made some changes, but more subtle. Or rather, a less-fleshed out Uma has their character traits tweaked or altered in their bios before implementation. Most notable case is probably Air Groove, with her "man hater" trait retconned away.
Also some characters like Oguri are slightly different between their anime/manga/game incarnations (though for Oguri the fanbase mixes all of them together since they don't really contradict).

Interestingly for Air Groove, her "main hater" trait in her original bio was due to a real life incident with the real Air Groove horse. But it would be pretty difficult to implement in-game since you can choose your trainer's gender and it'd make zero sense for Air Groove to be with a male trainer if she hated men, so that was dropped.
In terms of character changes between mediums, I think Mihono Bourbon had the biggest change, as her anime incarnation honestly acts quite different from how her in-game version acts.

That infamous incident involving Fine Motion's brother, right? Think they still adapted that somewhat by making Fine Motion's family give a pile of (room-hogging) gifts that Air Groove does not appreciate.
Heard also something about the real life horse not liking male handlers, but I am unable to substantiate the latter.
